概述 肠是从胃部到肛门的长而连续的管,肠系膜是将肠附着于肠壁的组织皱襞,肠和肠系膜结构非常不同,然而从蛋白质提取的角度来看,它们有一个共同点,它们很坚硬,难匀浆化,也很难进行总蛋白提取。使用传统的基于溶液法的蛋白质提取方法如RIPA,提取蛋白效率低、产率低,且提取的蛋白质的图谱不完整。Invent Minute试剂盒利用离心管技术以及优化的裂解液,通过物理研磨和化学裂解,可简单、快速地从人或动物的肠和肠系膜组织中高效提取蛋白质,并且可选择变性细胞裂解缓冲液或天然细胞裂解缓冲液以用于下游应用。本试剂盒整个操作过程不到10min,蛋白质收率在1~3mg/mL,足以进行50个样品的提取。

概述 脂肪组织特别是白色脂肪组织(WAT)已被证实除了具有能量储存功能外,还与内分泌和炎症有关。从脂肪组织中提取和分析蛋白质对于了解许多生理/病理状态越来越重要。但由于白色脂肪组织(WAT)和棕色脂肪组织(BAT)中高脂肪和低蛋白含量,在提取技术上更具挑战性。目前生物样品中水油乳化物非常难分离,Invent Minute具有特殊表面性质的带孔径的离心柱和优化的无表面活性剂的缓冲液系统,利用提取缓冲液比脂肪组织中油的冰点低,可将脂肪组织匀浆通过离心柱把水相和油相迅速分开,而组织中的总蛋白不丢失,蛋白质得率可达2~3mg/mL。

概述 Invent Minute质膜/蛋白和细胞组分分离试剂盒,通过离心柱技术配合特定缓冲液,可以快速从动物细胞或组织中分离天然的细胞膜膜片或膜蛋白。原理是细胞/组织通过Buffer A致敏,然后细胞以Z字形路径通过离心柱,在这个过程中细胞膜破裂,分离出完整的细胞核,获得的膜蛋白中不含有核膜和核蛋白污染。Invent试剂盒通过差速离心和密度离心可以把细胞蛋白分为总膜、细胞核、细胞质、细胞器及质膜,而无需超速离心,完成整个操作过程不超过45min。提示:本试剂盒最终获取的为细胞膜囊泡沉淀,如需进行蛋白实验,需使用膜蛋白溶解液溶解后,再进行下游实验。根据不同下游实验推荐使用货号WA-009(WB)、WA-010(IP、Co-IP)、WA-011(质谱)溶解液。

概述 植物膜蛋白占植物细胞总蛋白的很小一部分,但在植物生理学中起着非常重要的作用。传统的植物膜蛋白分离纯化方法是蔗糖密度梯度离心法和双液相法。这些方法虽然有效,但需要超速离心和大量的起始原材料,操作过程十分繁琐和费时。Invent Minute试剂盒解决了植物膜蛋白提取中的难点,使用时植物组织首先通过缓冲液A致敏,匀浆,然后通过离心柱,在此过程中,匀浆的组织通过柱子特有的Z字形通路后,细胞膜被切割成大小相等的碎片,后续无需超速离心,仅需通过差速离心和密度离心,可将天然的质膜蛋白从未破裂的细胞、细胞核、细胞质和细胞器的混合物中分离出来。在每次实验中仅需使用相同量的起始材料、离心力和离心时间,即可高度富集膜蛋白,并呈现高一致性。整个操作过程大约1小时。
